Study Reveals Why Aging Impairs Immune Response

Researchers discover metabolic changes that hinder disease treatment in the elderly says Kumar Chellappan

There is hope for old-age people suffering from certain ailments from which they had no route to escape hitherto. All treatments had failed to resuscitate them from the octopus-like shackles of the disease. A team of medical and bio-engineering researchers in the University Of Miami Miller School Of Medicine Immunology in the US have found out the reasons behind old men failing to respond to any treatments for a series of diseases. Nay, they have not invented any medicine for such diseases but have made a landmark invention about the reason behind the failure of drugs to cure the medical problems faced by the veterans.

A research paper published in Nature Communications, a widely followed scientific publication after peer review lists the reasons for the failure of medicines among old people. What makes the findings distinct is the part played by Dhananjay Suresh, a 20-year-old bio-medical engineer, and a key team member.

Dhananjay is waiting for admission to a leading medical school in the US for his MBBS course after the four-and-a-half-year-long B Tech course when he joined the team. Suresh is the son of India-born medical microbiologist couple Dr. Suresh Pallikuthu and Dr Ranjini Valiathan.

The researchers led by Dr. Daniela Frasca, principal investigator and associate professor of microbiology and immunology, have successfully proved for the first time in medical history that the aged fails to respond to medicines because of the “inability of immune cells to generate protective immunity”.

The metabolic abnormalities in immune cells lead to a reduced ability to generate protective immunity, according to Dr Frasca. Her team identified metabolic changes that decrease the body’s immunity as people age.

Dr Frasca explained that aging is associated with elevated levels of inflammation and metabolic changes. “We have identified and characterized age-associated metabolic changes that may accelerate and exacerbate deficiencies in immune cells from older adults, particularly those responsible for diminished secretion of protective antibodies against infections and vaccines.  

The research team also pinpointed the role a metabolic compound – long thought to be inessential – plays in immunity. Contrary to accepted wisdom, lactate can induce pathogenic B lymphocyte behavior and increase bodily inflammation.

“The metabolite lactate, the end product of anaerobic glycolysis, which has long been considered a discarded metabolic compound, plays a crucial role in age-associated defects in dysfunctional humoral responses,” Dr. Frasca said. “The effects of lactate, as described in the paper, recapitulate key features of B lymphocytes that infiltrate inflammatory tissues, where lactate-induced signaling may contribute to various pathological features.” Lactate can induce pathogenic B lymphocyte behavior and increase bodily inflammation.

With the script ready, medical doctors can step in and start a new therapy that will spur helpful immune responses in the elderly and keep the diseases at bay.

Now, it is for medical doctors to shape new therapies that selectively target metabolic products involved in the generation of inflammatory pathogenic immune cells to improve antibody responses in elderly individuals who are susceptible to infections and/or do not respond adequately to vaccination.

If the initial response from the world of medicine is any indication, this landmark research will go a long way in helping the aged to remain healthy and guarded against any kind of immune deficiency.
